Design and evolution of an enzyme with a non-canonical organocatalytic mechanism

A hydrolytic enzyme with a non-canonical organocatalytic mechanism was generated by introducing Nδ-methylhistidine through the cellular translation machinery into a designed active site, allowing optimization of enzyme performance using laboratory evolution protocols adapted to an expanded genetic code.
